There are two genes in the human genome that encode DIS3 proteins: hDIS3 and hDIS3L - they both interact andcooperate with the human core exosome. They have different intracellular localization patterns:

  • hDIS3 is mainly nuclear
  • hDIS3L is strictly cytoplasmic
Biochemical analyses indicate that both proteins are active 3' - 5' exonucleases but only hDIS3 displays endonucleolytic activity, which is in agreement with the preservation of the PIN active site in hDIS3 but not in hDIS3L. These results suggest that in humans the exosome complex endowed with endonucleolytic activity is mainly nuclear.
