tRNAs are transcribed by RNA polymerase III as pre-tRNAs in the nucleus.

tRNA transcription and end-processing occur in the nucleolus. Following end-processing [5' end processing - PMID: 16595295, 3' end processing - PMID: 18456844, 16336119, 19042972], CCA addition [PMID: 8617732], and various modification steps in the nucleoplasm, intron-containing pre-tRNAs are exported to the cytoplasm via the Los1 exportin and at least one unknown pathway [PMID: 3031485]. After pre-tRNA splicing on the cytoplasmic surface of mitochondria [PMID: 12925762, 17352735], additional modifications in the cytoplasm, and aminoacylation, mature charged tRNAs can participate in protein synthesis.
