Like its E. coli ortholog, T. thermophilus RsmB catalyzes the site-specific formation of m5C at position 967 (E. coli numbering) in the loop of helix 31 in 16S rRNA (adjacent to m2G966 catalyzed by RsmD). AdoMet is the methyl group donor. Together with the multi-site specific T. thermophilus RsmF, all four 5-methylcytidine (m5C) modifications are synthesized in T. thermophilus 16S rRNA. RsmB belongs to the same methyltransferase family as E.coli RsmF, yeast Nop2, yeast and archaeal Trm4, and homo sapiens P120, NSUN1, NSUN2, NSUN3 and NSUN4.
